1001 Gut bacterial sphingolipid production regulates the skin microbiome, skin lipid production and skin barrier homeostasis

چکیده
The stratum corneum serves as a critical barrier between the host and its external environment. Rich in ceramides, is one of largest sphingolipid reservoirs body. Our recent work using bioorthogonal labeling strategy traced sphingolipids from gut commensal Bacteroides thetaiotaomicron (BT) to liver mice fed with fat-free/high-sucrose diet demonstrated direct transfer labeled BT-sphingolipid liver. Additionally, synthesis ameliorated fatty-liver phenotype caused by high-sucrose diet. Currently, influence production on more distant sites, such skin are undefined, molecular mechanisms that mediate these interactions. Herein, we investigated impact transcriptome, concentrations, microbiota data show dysregulated lipid metabolism loss subcutaneous adipose layer induced fat-free were mitigated supplementing sphingolipid-producing B. thetaiotaomicron. regulated genes involved epidermal water balance, differentiation development tightly connected integrity function. Further, our demonstrate distinct community compared standard chow diet, expansions Staphylococcus Pseudomonas spp. addition BT-sphingolipids was able rescue phenotype. Taken together, findings importance phenotypes. also commensals metabolism, transcription, microbiota.

The lipid organisation in the skin barrier.
The main function of the skin is to protect the body against exogenous substances. The skin barrier is located in the outermost layer of the skin, the stratum corneum. This layer consists of keratin enriched cells embedded in lipid lamellae. These lamellae form the main barrier for diffusion of substances through the skin. In diseased skin the barrier function is often impaired. For a full unde...
متن کاملThe skin barrier from a lipid perspective.
This contribution summarises the results from a number of investigations undertaken in the spirit of the Domain Mosaic Model proposed by Forslind in 1994. Atomic Force Microscopy (AFM) studies on the two-dimensional phase behaviour of some stratum corneum lipids revealed phase separation of the lipids in the typical case and the ability of cholesterol to reduce the line tension between phases. ...
متن کاملThe skin microbiome: impact of modern environments on skin ecology, barrier integrity, and systemic immune programming
Skin barrier structure and function is essential to human health. Hitherto unrecognized functions of epidermal keratinocytes show that the skin plays an important role in adapting whole-body physiology to changing environments, including the capacity to produce a wide variety of hormones, neurotransmitters and cytokine that can potentially influence whole-body states, and quite possibly, even e...
متن کاملIL-4 regulates skin homeostasis and the predisposition toward allergic skin inflammation.
IL-4 promotes the development of Th2 cells and allergic inflammation. In atopic dermatitis lesions, IL-4 decreases the expression of multiple genes associated with innate defense, including genes in the epidermal differentiation complex (EDC) that regulate epidermal barrier function. However, it is not clear whether IL-4 also contributes to homeostatic control of EDC genes. In this report, we d...
متن کاملUrban particulate matter down-regulates filaggrin via COX2 expression/PGE2 production leading to skin barrier dysfunction
We explored the regulation of filaggrin, cyclooxygenase 2 (COX2) and prostaglandin E2 (PGE2) expression induced by urban particulate matter (PM) in human keratinocytes. In addition, we investigated the signaling pathways involved in PM-induced effects on COX2/PGE2 and filaggrin. PMs induced increases in COX2 expression and PGE2 production, and decreased filaggrin expression. These effects were ...
